ENSAYO CLÍNICO

Objectius del projecte

Objetivo Principal: ParteA •Determinar seguridad y tolerabilidad de CC-90010+ TMZ como tratamiento adyuvante •Determinar seguridad y tolerabilidad de CC-90010+TMZ+RT como tratamiento concomitante •Definir dosis máxima tolerada (DMT) y/o la dosis recomendada de fase 2(DRP2) de CC-90010+TMZ como tratamiento adyuvante •Definir DMT y/o DRP2 de CC-90010 en combinación con TMZ+RT como tratamiento concomitante ParteB •Evaluar la eficacia de la combinación CC-90010+TMZ con RT concomitante y CC-90010+TMZ como tratamiento adyuvante seguido de CC-90010 en monoterapia en -sujetos con GBM de nuevo diagnóstico(Brazo A) frente al brazo estándar de TMZ con RT concomitante y tratamiento adyuvante con TMZ(Brazo B) •Evaluar seguridad y tolerabilidad de la combinación de CC-90010 + TMZ con RT concomitante y CC-90010 + TMZ como tratamiento adyuvante seguido de CC-90010 en monoterapia(Brazo A) frente al brazo estándar de TMZ con RT concomitante y tratamiento adyuvante con TMZ(Brazo B) Objetivos Secundarios: Parte A ? Evaluar la eficacia preliminar de la combinación de CC-90010 + TMZ con RT concomitante, y como tratamiento adyuvante en sujetos con GBM de nuevo diagnóstico ? Caracterizar la farmacocinética (FC) de CC-90010 administrado en combinación con TMZ como tratamiento adyuvante y con TMZ + RT como tratamiento concomitante. Parte B ? Comparar la SLP y la SG de la combinación de CC-90010 + TMZ con RT concomitante y CC-90010 + TMZ como tratamiento adyuvante seguido de CC-90010 en monoterapia (Brazo A) frente al brazo estándar de TMZ con RT concomitante y tratamiento adyuvante con TMZ (Brazo B), mediante la estimación de lo siguiente en cada brazo terapéutico: – Tasas de SLP a los 6 y 12 meses – Tasas de SG a los 6 y 12 meses – Mediana de la supervivencia global(mSG) – Duración del tratamiento(DdT) ? Caracterizar más a fondo la FC de CC-90010 administrado con TMZ + RT como tratamiento concomitante en combinación con TMZ como terapia adyuvante en el Brazo A.
